Did you install the asmail port or did you get asmail from the
afterstep-devel port?  If you used the asmail port/package there should be
a sample .asmailrc in /usr/X11R6/share/doc/asmail/sample.asmailrc.

Where are you trying to start asmail?  From a command line or inside your
.steprc (or inside the wharf file for afterstep-1.4.5.3 - the latest
port)?

Here's what mine looks like.  

	MailFiles
	        /home/brett/mail/mbox
	End
