Which program are you using to play the audio file? If you're using XMMS in GNOME, right-click on the XMMS window (somewhere with blank space, like between the "Eject" and "Rand" buttons), choose "Options" then "Preferences". In the new window, make sure that "eSound Output Plugin" is selected as the output plugin.
