You won't be able to unmount a partition that is in use:

[root@mir root]# umount /usr
umount: unmount of /usr failed: Device busy

AFAIK the only way to do this is to put the tunefs lines at the
appropriate point in /etc/ec then reboot. Search the list archives for
the exact place in /etc/rc, I don't remember off the top of my head.
