My concern is with reading. Maildirs are totally safe for reading or writing via NFS (or anything else) with or without locking, including environments with multiple servers doing deliveries. You would have to use a POP server that supports maildirs (included with qmail) or switch to a reader that supports them (ie: MUTT).
