I have recently switched to 10-STABLE and WITH_NEW_XORG, removed all ports and installed needed ports from scratch, including mono. Then tried to use mono to start an application (KeePass.exe) which failed complaining about not being able to load libgdiplus. Installing the x11-toolkits/libgdiplus port fixed this problem, so I assume a dependency is missing here.

Install Mono port on empty machine including all dependencies and try to start mono with a program using a GUI.
