It seems (from my perusal of the man pages) that there is no provision
to run lukemftpd in standalone mode, ie. without inetd, if that's what
you mean.
So, just edit /etc/inetd.conf and uncomment the lukemftpd line.
Comment out any reference to the standard ftpd if they're enabled.
Then send a HUP signal to the inetd process:
kill -HUP `cat /var/run/inetd.pid`
