Have a look at /etc/make.conf, especially MASTER_SITE_OVERRIDE and MASTER_SITE_BACKUP. You cannot override port-defined donwlod sites, though. If you got constant difficulties downloading port tarballs, have a look at the filename and download from your preferred mirror to /usr/ports/disfiles by hand.
