After investigation it turns out it requires an helper program it expects to find in PREFIX/lib/virtualbox/VBoxNetNAT which is built but not installed by the port. I made a simple patch fixing this issue. Also the virtualbox 5 port I found at [1] needs the same fix. [1] https://people.freebsd.org/~jkim/vbox/help
